Research Areas:
My scientific interest is on understanding the molecular basis of environmental perception of cell membrane and cross-membrane signal transduction. The research focuses on the interaction between lipid molecules and protein receptors on eukaryotic cell membranes. The classic theory of cell membrane receptor activation is based on ligand-receptor recognition, which centralizes protein-protein interactions only. However, an increasing amount of experimental data confirms that receptors can spontaneously undergo activation in the absence of ligands. This none ligand-dependent receptor activation typically occurs when the lipid microenvironment undergoes changes, indicating that lipid molecules play an important role in receptor activation. Therefore, the classic protein centered receptor activation theory needs to be improved and modified, and the interaction between proteins and lipid molecules should be included in the theory of receptor activation. With the help of advanced biophysical methods, on the one hand, the aggregation and movement status of receptors can be characterized in situ at the single-molecule level, and on the other hand, the purified transmembrane protein can be reconstructed in lipid micelle in vitro, and the influence of lipid composition on the structure of receptor transmembrane can be characterized by liquid nuclear magnetic resonance. Ultimately, we hope to gain a comprehensive understanding of the receptor activation on eukaryotic cell membranes.
